    C57BL/6 mice were either home-bred or were purchased from SamTako Ltd (South Korea). B6.SJL, Pmel Tg , Il17a Cre mice were purchased from Jackson Laboratory. Foxp3 DTR mice were kindly provided by Dr. Sin-Hyeog Im (POSTECH) with the permission of Dr. Alexander Rudensky (Memorial Sloan Kettering Cancer Center).23 R26 DTA mice and R26 YFP mice were previously described.24 25 Il17a Cre ×R26 YFP , Il17a Cre ×R26 DTA, and Il17a Cre ×R26 DTA/YFP mouse systems were created by breeding the relevant strains.     Mice used included C57BL/6J (Charles River, #027), Tlr4 KO (Jackson Laboratory, #029015), Tlr4 floxed (Jackson Laboratory, #024872), iDTR and Mcpt5-Cre + (provided by Axel Roers, University of Technology, Dresden, Germany). All mice were used between 6 and 10 weeks. For mast cell depletion in Mcpt5-Cre + x iDTR mice, 8-week old mice were given 200 ng Diphtheria Toxin (DT, Unnicked, from Corynebacterium diphtheriae, List Biological Laboratories) per mouse intravenously once a day for 5 consecutive days prior to intradermal ear immunization.     Homozygous PANIC-ATTAC mice were administered a single dose of AP20187 homodimerizer (0.08125 mg/kg) prepared according to the manufacturer’s instructions (17). Female NOD mice were surveyed twice-weekly by glucometer, until spontaneous diabetes was diagnosed by glucose >250 mg/dL for two consecutive measurements. DT (List Biological Laboratories; 150) was administered to NSG-DTR mice in a single, 300-μL intraperitoneal injection of 5.0 ng total DT. Control NSG-DTR mice (phosphate-buffered saline; PBS) were treated with an equal volume of 1× PBS (Sigma-Aldrich). All animals in a cohort (with human islets from the same human donor) were injected with DT or PBS on the same day (29).

  • 2.5. Multiplex immunoassay

    IgG concentrations specific to vaccine antigens PT, PRN, FHA, TT and DT were measured using an in-house multiplex bead-based immunoassay (MIA). IgG specific to FIM 2/3, which is not included in Boostrix, was also measured as a proxy for natural infection. PT, PRN and FHA were kindly provided by GlaxoSmithKline (Belgium). TT was purchased from Sigma-Aldrich. DT and FIM 2/3 were sourced from List Biological Laboratories (USA). A standard curve was generated using a 10-step 3-fold serial dilution of an in-house reference serum. Blanks and two quality control (QC) samples were included on every plate to calculate % critical variance across all assays, which fell between 5.1 and 14.6%.
