Animal Models (Cell depletion):
The CD11b+ cells were depleted in CD11b-diphtheria toxin receptor (DTR) mice (congenic in the C57BL/6 background) by intraperitoneal injection with diphtheria toxin (DT; 7.5 ng/g body weight; List Biological Laboratories) or phosphate-buffered saline (PBS) on days 7 and 9, with sacrifice on day 10. We gave DT twice as continued DT treatment is toxic and can lead to saturation of the DTR. Effects of CD11b+ cell depletion were assessed on day 8 and day 10 (1 day after treatment) as CD11b+ cells repopulate the liver 48 hours after DT depletion.
