  • Induction of experimental autoimmune orchitis (EAO)

Age-matched male Gsdmdfl/flCx3cr1-cre and Cx3cr1-cre mice were used to induce EAO, as described previously (Nicolas et al, 2017). First, the testicular homogenate (TH) was obtained from decapsulated testes of adult syngeneic mice and homogenized in an equal volume of sterile PBS. Second, TH was mixed with incomplete Freund’s adjuvant (IFA; Sigma-Aldrich, F5506) containing 1 mg/ml Mycobacterium tuberculosis (BD, 231141) in 1:1 ratio. Finally, the mice were immunized thrice every 14 days with the mixture, followed by an intraperitoneal injection of 100 ng pertussis toxin (List Biological Laboratories, lot#180) in 100 μl PBS. Each animal was immunized dorsally at four sites near the popliteal lymph nodes in a total volume of 200 μl. The testes were collected for further analysis 50 days after the first immunization.

  • USA). MOG35-55 (2 mg/mL in TDW) was vigorously mixed with the same volume of CFA using a homogenizer (Qiagen). The peptide emulsion was injected subcutaneously at 200 ?g per mouse. Pertussis toxin (PTX; List Biological Labs, Campbell, CA, USA) was prepared in PBS and intraperitoneally injected at 400 ng per mouse, at 2 hr and 24 hr post administration of the CFA emulsion. For the prophylactic treatment, the nanoparticles were administered subcutaneously at 1 mg of PN and 6 ?g of dexamethasone per mouse at 1 and 2 weeks before injection with the MOG/CFA emulsion.
