Induction and assessment of EAE

This modeling method is based on previous research [37]. Eight-week-old female C57BL/6 mice were injected subcutaneously in the dorsal flanks with 150 μg MOG35-55 peptide (RS Synthesis) emulsified in CFA (5 μg/mL). Pertussis toxin (500 ng per mouse, list labs) was given i.v. 24 h later. A second MOG and pertussis injection was given 7 d later. LND was intraperitoneally injected daily (60 mg/kg) beginning at disease induction. Mice in the vehicle group were injected with β-CD at the same time points. Animals were monitored and scored daily until they were euthanized. Clinical scores were assessed as follows: 0 = no symptoms, 1 = loss of tail tonicity, 2 = unsteady gait, 3 = hind limb paralysis, 4 = forelimb paralysis, 5 = moribund or dead. If scores fell between two points, 0.5 gradations were used.

EAE induction and clinical scoring

Two weeks post-AAV vector delivery, EAE was induced in C57BL/6J female mice as previously described.8 Emulsion containing 150 μg of MOG33-55 (AnaSpec) in complete Freund’s adjuvant (CFA) (MilliporeSigma) with 600 μg of Mycobacterium tuberculosis (BD) was subcutaneously injected into both sides of the lateral abdomen. A 375 ng of pertussis toxin (List Biologicals) was injected intraperitoneally on days 0 and 2. For healthy controls, the same emulsion without MOG33-55 peptide was used. EAE clinical symptoms were monitored on a daily basis following Hooke lab’s Mouse EAE scoring guidelines.

Peanut sensitization, ButM treatment and challenge

SPF C3H/HeN mice were treated with 0.45 mg of vancomycin in 0.1 ml volume by intragastric gavage for 7 d pre-weaning and then with 200 mg l−1 vancomycin in the drinking water throughout the remainder of the sensitization protocol. Age- and sex-matched 3-week-old littermates were sensitized weekly by intragastric gavage with defatted, in-house-made peanut extract prepared from unsalted roasted peanuts (Hampton Farms, Severn, NC) and cholera toxin (CT) (List Biologicals) as previously described25,45. 2.3. EAE induction and evaluation

EAE was induced using methods in a previous study [23]. Briefly, 200 μg of murine myelin oligodendrocyte glycoprotein (MOG) (GL Biochem, Shanghai, China) was emulsified in complete Freund’s adjuvant (CFA) [18], which consisted of 5 mg/mL Mycobacterium tuberculosis (BD, San Jose, CA, USA) and incomplete Freund’s adjuvant (Sigma-Aldrich, St. Louis, MO, USA). After which, the mice were injected with MOG/CFA subcutaneously on the left and right retroauricular spots and flank on day 0.
