Product: Anti-Cholera Toxin B Subunit (Goat)

  • … For Fig. 2B, we performed double fluorescent labelling of immunoreactivity for CTB and mRNA for VGluT1 or GAD65/67 as follows. … To amplify the DNP signals, the sections were washed in TNT three times for 10 min each, incubated with an anti-DNP antibody conjugated with horseradish peroxidase (1:500; Perkin-Elmer) for 1 h at room temperature, and treated again with diluted TSA-Plus DNP reagents (1:100) for 5 min. Subsequently, the sections were incubated overnight with an anti-DNP antibody conjugated with Alexa 488 (1:500; Molecular Probes) in 1% blocking buffer at 4C for fluorescence detection of DNP signals. At this point, a goat anti-CTB antibody (1:500; List Biological Laboratories #703) was added to the incubation mixture for detection of CTB. …

Product: Pertussis Toxin from B. pertussis, Lyophilized (Salt-Free)

  • MOG immunization and EAE:

Mice were immunized with 100 mg of myelin oligodendrocyte glycoprotein (MOG)3555 peptide in CFA (Difco) with 500 mg of Mycobacterium tuberculosis (Difco). Mice were i.p. injected 200 ng of pertussis toxin (List Biological,#181) on the day of immunization and 2 days later, as described (Kang et al., 2010). The mice were observed daily for clinical signs and scored as described previously (Shi et al., 2011).

Product: ULTRA PURE LPS from Escherichia coli O111:B4

  • Reagents:

Ultrapure LPS (Escherichia coli 0111: B4) was purchased from List Biological Laboratories (Vandell Way, CA, USA). This LPS does not contain contaminating proteins that could stimulate TLR2 nonspecifically. …

    Experimental model:

C57BL/6 male mice at 8-10 weeks of age were used for the present experiments. Mice were housed in a temperature- and humidity-controlled environment on a constant 12:12-h light-dark cycle. All animal studies were approved by the Animal Ethics Committee of Jinzhou Medical University. Mice were fasted for 12 h with water ad libitum before the operation. Mice were anesthetized with 2% isoflurane, and common BDL was performed as previously described [26]. In brief, following a peritoneal cavity incision, the common bile duct was dissected and double-ligated with a 5-0 silk suture, and then the bile duct was cut between the ligatures. Controls underwent a sham operation in which the common bile duct was exposed but not ligated. A two-layer running suture was used for abdominal closure with 4-0 dexon and 2-0 nylon. Two concentrations of DA (25 or 50 mg/kg) were administered intraperitoneally daily at 9:00 AM beginning after the ligature on day 1. Four days after BDL, selected mice were subjected to an ileocecal vein injection of LPS under microscopy at a dose of 100 ng/ gram of body weight (gbw), 1 g/gbw or 5 g/gbw diluted with 300 l of PBS.
