  • Tracing of myelinated afferent fiber projections to the dorsal horn:

The transganglionic tracer, cholera toxin B-subunit (CTB), has been used to map the central projections of cutaneous myelin primary aerents in the spinal dorsal horn of rats.25 After six weeks of RTX injection, CTB (1%, List Biological Laboratories, Campbell, CA) was injected into left sciatic nerve of vehicle-, RTX-, RTXMDL28170-, and RTXPEG400/DMSO-treated rats to determine the eect of MDL28170 on RTXinduced sprouting of myelinated aerent bers labeled by CTB in the dorsal horn. The sciatic nerve was exposed at the level after the rats were anesthetized with 10% chloralic hydras (3.5ml/kg, intraperitoneal). Four microliters of CTB tracers were loaded into Hamilton Yuan et al. 3 microsyringes and injected into the sciatic nerves on the left side of the body. …

Product: Epsilon Toxin from C. perfringens

  • Determination of mAb affinity by surface plasmon resonance:

The affinities of mAbs were determined by surface plasmon resonance (SPR) on a BIAcore T200 instrument (GE Healthcare, Uppsala, Sweden) by carrying out two independent multi-cycle kinetic measurements for each antibody. All analyses were performed at 25°C on a CM5 sensor chip in running buffer (10 mM HEPES, 150 mM NaCl, 3 mM EDTA, 0.05% Tween 20). Polyclonal anti-mouse IgG antibody was covalently conjugated to a CM5 sensor chip according to the manufacturers protocol (Mouse Antibody Capture Kit, GE Healthcare, Uppsala, Sweden). Individual mAbs (1 µg/mL) were bound to this mouse capture chip for 30 s at 5 µL/min. Each captured antibody was subjected to five consecutive threefold serial dilutions of epsilon toxin (ranging from 2.47 to 1800 nM; List Biological Laboratories) for 120 s at a constant flow rate of 30 µL/min to obtain a maximum signal of approx. 30 resonance units (RU). …
