    Cell preparation:

    RAW Cells were plated at 5x106cells in 5ml of RPMI media supplemented with 10%FCS and 1%P/S into 25cm2 flasks. At 70% confluence, 120M of BSO, an inhibitor of the synthesis of GSH, was added to the culture for overnight incubation. The next day, cells were stimulated for 2h with LPS. In total four experimental conditions were tested: control, LPS, BSO, BSO+LPS.

  • LPS administration:

    The TLR4 ligand LPS (E. coli 055:B5, Catalog no. 423, Lot no. 4231A1) was purchased from List Laboratories. Two doses of LPS (5 and 10 mg/kg, in sterile saline) were used to test the induction of white matter injury in WT mice as described in the Results. Saline-injected mice served as controls. For induction of sepsis, LPS doses were injected subcutaneously between the shoulders of PND 2 mice as previously described [9].
