2.2. Preparation of Antigen and Induction of EAAU
Melanin-associated antigen (MAA) was prepared according to the method proposed by Broekhuyse et al. [8, 11]. To induce EAAU, the rats were given two simultaneous injections of different MAA preparations. First, MAA was suspended in phosphate-buffered saline (PBS) and 1 : 1 emulsified in complete Freund’s adjuvant (Sigma Aldrich, St. Louis, MO, USA). The suspension (0.05 mL) was injected into the left hind footpad. Second, MAA was emulsified with 1 μg of purified Bordetella pertussis toxin (List Labs, Campbell, CA, USA) and injected into the peritoneal cavity for a total volume of 0.05 mL.

Oral DMF Gavage and EAE Induction
… Induction of active EAE was performed as previously described (38, 39). For DC transfer experiments, 1 × 105 DC, cultured in vitro for 10 days with or without 200 µM MMF (Sigma-Aldrich) and stimulated with 1 µg/ml lipopolysaccharide (LPS) (Sigma-Aldrich) for 24 h, were injected intraperitoneally at d2 and d7 after EAE induction together with 200 ng pertussis toxin (List Biological Laboratories).

Spin trapping coupled to EPR:
… This technique was used in this project to evaluate the ROS species release from cells after stimulation by LPS, and if this environment was modified by treatment with BSO. BMPO (5-tert-butoxycarbonyl 5-methyl-1-pyrroline N-oxide) was used as a spin trap. It specifically reacts with O2.- forming a superoxide adduct, BMPO-OOH, or with HO. forming the hydroxyl adduct, BMPO-OH which is characterised by a 4 lines absorbance spectrum (Figure 2.2). …
Cell preparation:
RAW Cells were plated at 5x106cells in 5ml of RPMI media supplemented with 10%FCS and 1%P/S into 25cm2 flasks. At 70% confluence, 120M of BSO, an inhibitor of the synthesis of GSH, was added to the culture for overnight incubation. The next day, cells were stimulated for 2h with LPS. In total four experimental conditions were tested: control, LPS, BSO, BSO+LPS.

Product: ULTRA PURE LPS from Escherichia coli O55:B5
LPS administration:
The TLR4 ligand LPS (E. coli 055:B5, Catalog no. 423, Lot no. 4231A1) was purchased from List Laboratories. Two doses of LPS (5 and 10 mg/kg, in sterile saline) were used to test the induction of white matter injury in WT mice as described in the Results. Saline-injected mice served as controls. For induction of sepsis, LPS doses were injected subcutaneously between the shoulders of PND 2 mice as previously described [9].
