Determination of mAb affinity by surface plasmon resonance:
The affinities of mAbs were determined by surface plasmon resonance (SPR) on a BIAcore T200 instrument (GE Healthcare, Uppsala, Sweden) by carrying out two independent multi-cycle kinetic measurements for each antibody. All analyses were performed at 25°C on a CM5 sensor chip in running buffer (10 mM HEPES, 150 mM NaCl, 3 mM EDTA, 0.05% Tween 20). Polyclonal anti-mouse IgG antibody was covalently conjugated to a CM5 sensor chip according to the manufacturers protocol (Mouse Antibody Capture Kit, GE Healthcare, Uppsala, Sweden).
