  • Oral immunization:

    Mice received sodium bicarbonate to neutralize gastric acid before oral immunization (9, 22). Thirty minutes later, the mice were immunized orally concurrently with 1 mg OVA (Sigma-Aldrich) and 10 mg cholera toxin (CT; List Biological Laboratories, Campbell, CA). This oral immunization procedure was conducted on days 0, 7, and 14.

  • Ovalbumin food allergy model:

    The OVA food allergy model has been described previously( 16 , 24 ). Briefly, 6-week-old conventional female BALB/c OlaHsd mice (10–15 animals per group) (Harlan Laboratories) were sensitised via the oral route daily during the first 3 d and then at weekly intervals with 20 mg of OVA (Sigma) along with 10 μg/mouse of cholera toxin used as an adjuvant (List Biological Laboratories) for a continuous period of 7 weeks. …

Product: ULTRA PURE LPS from Escherichia coli O111:B4

  • Analysis of T-cell and monocyte population activation:

    …Monocyte populations in the PBMC samples (200 l per well) were stimulated in separate ninety-six-well plates with 10 g/l of ultra-pure lipopolysaccharide (LPS; List Biological Laboratories, Inc.) per well or an equal volume of sterile endotoxin-free water as a negative control. PBMC were incubated at 37°C in 5 % CO2 and the supernatants were removed after 24, 48 and 72 h for the analysis of monocyte-derived cytokines as described below.

Bremer, PT;Hixon, MS;Janda, KD;

Product: SNAPtide® Peptide Substrate (FITC/DABCYL) for C. botulinum Type A Neurotoxin

  • SNAPtide Assay[44]

    Recombinant 425aa BoNT/A LC was used throughout all assays and was prepared as previously described.[45] All SNAPtide assays were run in 40 mM HEPES + 0.1% Triton X-100 at pH 7.4. BoNT/A LC concentrations were 18.5 nM while SNAPtide (prod. no. 521, List Labs) concentrations were 5 M. Fluorescence was recorded continuously for 1.5 h to calculate an accurate kobs using equation 1. In the preincubation experiments, inhibitors at the specified concentrations were incubated with 925 nM enzyme and diluted 50-fold into substrate and initial rates of SNAPtide cleavage were measured. …
