  • 2.1. Mouse model and treatment

    Vglut3DTR/+ mice were generated in our lab. We used the CRISPR/Cas9 approach to connect DTR to the C-terminus of Vglut3 via the P2A peptide. PCR and Southern blot methods were used to screen out the mice with the correct recombination. Lgr5CreERT2/+ (Stock #008875) and Rosa26-tdTomato (Stock #007914) mice were purchased from the Jackson Laboratory. The primers used for genotyping Lgr5creERT2/+, Rosa26-tdTomato and Vglut3DTR/+ mice are listed in Supplementary table 1. Tamoxifen (Sigma-Aldrich) was injected intraperitoneally at postnatal day (P)2 at 0.20 mg/g bodyweight. EdU (Click-iT EdU Imaging Kit; Invitrogen) was injected intraperitoneally from P3 to P5 once a day at a dose of 50 μg/g.   • Immunization of the replication cohorts followed the protocol of Wagnon et al. [17].

    Male C57B/6 J mice were immunized at 8–9 weeks of age with either GluN1168-187 (control peptide), GluN1359-378 (active peptide) or for our additional comparison with ovalbumin, each emulsified in an equal volume of CFA (as described above). Antigens (200 µg) were equally distributed over 4 subcutaneous injections, 2 at shoulders and 2 at hind limbs.     This single-center, randomized, double-blind, placebo-controlled, full-factorial single-dose study was conducted between December 2016 and December 2017 in subjects in whom pyrexia was induced by intravenous (IV) administration of a reference standard endotoxin (RSE; ClinicalTrials.gov identifier: NCT02761980).
