2. Signal evaluation by recognition of T. denticola via TLR2 or TLR4
... Escherichia coli O55:B5 LPS (List Biological Labs, Inc. Ca, USA) was added to TLR4-reporter cells for a positive control. In a preliminary experiment, the phosphatase activity of T. denticola was confirmed to be negligible (data not shown).
Subjects and experimental design
... After baseline (BL) measurements, animals were subjected to a continuous lipopolysaccharide (Escherichia coli O5:55, List Biologic Laboratories Inc, USA) infusion of 10 ng/kg/min for 20 h to induce lung injury16. Then measurements were repeated for lung injury (INJ). During the time of the experiment, PEEP and FiO2 were managed according to the ARDS Network table (low PEEP high FiO2)17.
Lipopolysaccharide murine acute lung injury model
... Eight- to ten-week-old C57BL/6 J female mice (Jackson Laboratory) were injured intratracheally (IT) with lipopolysaccharide (LPS, O55:B5 #203A, List Labs). Each mouse received 50 μg LPS dissolved in 50 μL sterile phosphate buffered saline (PBS) at time zero. Control mice were administered 50 μL IT PBS. ...
... 50 ng/mL rhM-CSF plus 10 ng/mL lipopolysaccharide from Escherichia coli O55:B5 (LPS; List Biological Laboratories, Campbell, CA, USA) for M1, ...
... Then, adherent macrophages underwent further incubation in serum-free RPMI1640 at 37°C for 2h and subsequent treatment with LPS (List Biological Laboratories, Escherichia coli 055:B5), ...
Experimental animal models
In the second series of experiments, mice, under light anesthesia, were instilled intranasally with 60 μg of lipopolysaccharide (LPS) (Escherichia coli 055:B5; List Biological Laboratories, Campbell, CA, USA) in 60 μl of sterile 0.9% NaCl solution. Control animals received equivalent volume of vehicle saline solution. When bevacizumab, which neutralizes VEGF and blocks its signal transduction through VEGF receptors (Papadopoulos et al. 2012), was used, it was intravenously given to mice at a dose of 40 μg at 60 min before administration of LPS. Animals were euthanized at 24 h after LPS challenge. All experimental data were analyzed in a blinded fashion.
... For macrophage activation, the adherent cells were stimulated with 1g/mL of lipopolysaccharide from E.coli O55:BS (List Biological Lab Inc.) in media for 4 h at 37 C ...
