    To detect PT production by Western blot, to a 1.0 mL aliquot of culture supernatant, ¼ volume of 100% TCA was added, mixed, and the sample was held on ice for 1 h. Samples were centrifuged at 16,000 × g for 30 min at 4 °C and the supernatant was removed.     The assay was run using cryopreserved human PBMCs from a single healthy donor and were allowed to recover for 24 hours before use.
