    Breeding cages were randomly assigned to one of the experimental isocaloric diets (AIN-93G, BioServ): standard (Stand, BioServ #S7628), n-3 long chain (LC)-PUFA enriched (n-3, BioServ #S7630; soybean oil replaced with Menhaden Fish oil) or n-6 LC-PUFA supplemented (n-6, BioServ #S7629; based on corn oil). The detailed composition of the diets is presented in our previous publication [14]. The day of birth was defined as postnatal day (P) 0. At P9, pups received a single intraperitoneal injection of lipopolysaccharide (LPS from E. coli O55:B5, 1 mg/kg, Cat# 423, List labs) or equivalent volume of saline. The dams remained on the experimental diet until offspring sacrifice (P10-P12). To allocate pups to particular experimental groups (LPS or saline), randomization in matched pairs was performed based on sex and weight of animals [15]. Number of animals in each group are shown in Suppl. Table 1.

    Overlapping peptide pools targeting ESAT6 and CFP10 were used to stimulate T cells for these studies (BEI Resources). Peptides were 15 or 16 mers with 11 or 12 amino acid overlaps for ESAT6 protein and 11 amino acid overlaps for CFP10. Mtb whole-cell lysate from H37Rv was also used to stimulate T cells (BEI Resources). Dimethyl sulfoxide (DMSO) (Sigma-Aldrich) was used as a negative control. Staphylococcus enterotoxin B (List Biological Laboratories) was used as a positive control for the low-exposure cohort.
