Cell culture and reagents:
A spontaneously transformed type II mouse lung alveolar epithelial cell line [19] (C10) was cultured as described previously [20]. A C10 cell line stably expressing NF-B luciferase was generated as previously described [21]. Briefly, C10 cells were transfected with the plasmid, 6 B-tk-luc, in the presence of the plasmid PSV-2Neo (Promega), conferring a resistance to neomycin. Following incubation with the antibiotic, geneticin, antibiotic-resistant colonies were propagated. C10 cells were starved in medium containing 0.5% FBS for 16 h prior to exposure of 1 g/mL LPS (List Biological Laboratories, Inc).

Human Monocyte-Derived Macrophages (MDM) and MSC Direct Coculture:
Before each experiment, MSC were trypsinized, counted, washed with sterile 1X DPBS, resuspended in RPMI-1640 medium 1% FCS and added to the culture of primary human macrophages at a 1:20 MSC/MDM ratio. Cells were stimulated with LPS (Escherichia coli O111:B4, List Biological Laboratories (Campbell, California, www.listlabstage.wpengine.com) 10 ng/ml).
