Product: LPS from Escherichia coli O55:B5
Prostaglandin E2 measurement:
Blood samples were transported on ice to the lab within one hour. The samples were aliquoted into sterile polypropylene tubes and were incubated at 37C on an orbital roller in the absence (control) or presence of O55:B5 LPSb at a final concentration of 100 g/ml. After 6 h, the samples were centrifuged, 100 L of the plasma was combined with 900 L of methanol and dried by evaporation. …

Product: Anthrax Edema Factor (EF), Recombinant from B. anthracis
In vitro protease assays:
Purified InhA1 was mixed with the substrate in protease buffer (50 mM Tris, 100 mM NaCl, 1 mM CaCl2, 1 mM MgSO4 [pH 7.8]) and incubated at 37C for set time points. Reaction mixtures were then placed on ice, and EDTA was added to a final concentration of 50 mM to stop the reaction. SDS buffer (2) was added, and samples were boiled for 5 min, separated by SDS-PAGE, and analyzed by Western blotting with substrate-specific antibodies. Recombinant PA (rPA), rLF, and rSodA-1 from B. anthracis were obtained from BEI Resources (ATCC, Manassas, VA). rEF was obtained from List Biological Laboratories (Campbell, CA). rNpr599 was purified as described above.

HTLV-1 p30 decreases TLR4 expression:
… These data are consistent with the finding of Datta and colleagues (3) which
demonstrated that p30 decreases the release of pro-inflammatory cytokines, such as TNF-g, IL-8,
and MCP1 upon stimulation with the TLR4 specific agonist lipopolysaccharide (LPS) by
decreasing the expression of TLR4 itself. Similarly, we transduced THP-1 cells with p30
lentivirus or with the control lentivirus and after 12 hours of LPS stimulation, cells were
collected and analyzed by flow cytometry for intracellular production of IL-12 and TNF-g (Fig. 241
1C). In both cases, the presence of p30 decreased the expression of such cytokines upon LPS
stimulation when compared with the control.
