  • Animals and deafening protocol

Diptheria toxin (DT; List Biologicals Lab. Inc. cat. #150) was dissolved in saline, aliquoted and stored at − 20 °C until use. Repeat freeze–thaw cycles were avoided. A single dose of 20 ng/g DT was administered intra-muscularly to adult mice. Mice receiving adenovirus (Ad) inoculation surgery, were given DT immediately after the surgery. High-calorie diet gel (DietGel 76A, ClearH20) and an extra bottle of water were provided for the 2 following weeks. Body weight and general health metrics were monitored every day for a week after DT administration.

    Tumor Associated Macrophage Co-Injection Experiments

Foxp3DTR-GFP donor mice were injected as mentioned above with either PyMT or EO771 cells, and their tumors were allowed to grow until approximately 90mm3. Half of the donors received two doses of 50 μg/kg of diphtheria toxin (DT) in two consecutive days, and primary tumors from both groups were harvested four days after initial DT treatment.

  • Mice and reagents.

C57BL/6 (B6), B6 CD45.1, BALB/c, CBA, B6 Foxp3-DTR, and B6 Foxp3-IRES-GFP mice were purchased from The Jackson Laboratory. As indicated, select lung and heart transplant recipients were treated with perioperative costimulatory blockade consisting of anti-CD40 ligand (250 μg intraperitoneally [i.p.], day 0) and CTLA-4–Ig (200 μg i.p., day 2) (Bio X Cell). Diphtheria toxin (DT) (List Biological Laboratories Inc.) (500 ng i.p. days 0, 1, and 7) was given to select B6 Foxp3-DTR heart transplant recipients to deplete Foxp3+ cells. …
