Product: Diphtheria Toxin, Unnicked, from Corynebacterium diphtheriae

  • CD115+ M-MDSC depletion

MMDTR mice were implanted with 2.5 × 105 EL4 cells. Each mouse received one intraperitoneal injection (10 ng/g of body weight) of diphtheria toxin (DT) (List Biological Laboratories, Campbell, CA, USA) on days 7, 9, and 11 after tumor implantation (Supplementary Fig. 7b). Time intervals (every 48 h) of DT injection were optimized by flow cytometry blood analysis (Supplementary Fig. 7a). After sacrifice on day 14 after tumor implantation, single cells were prepared for flow cytometry analysis. CD11b+Ly6G+Ly6Clo cells from naive or TB mice were sorted for OLFM4 immunohistochemistry (IHC) and qRT-PCR analysis. C57BL/6 mice were used as a negative control to exclude effects related to DT.

  • Mouse techniques

… For diphtheria toxin receptor (DTR) ablation studies, WT and indicated DTR mice received injection twice intratracheally (i.t.) with 100 ng diphtheria toxin (DT) in 20 μl of sterile PBS or i.p. with 200 ng DT in 100 μl of sterile PBS (150, List Biological Lab) on 1 d before and after metastatic inoculation. Cell of interest depletion was confirmed by flow cytometry. …

  • Tregs depletion and agent administration

To deplete Tregs, DEREG mice were injected intraperitoneally with 1 μg DT (List Biological Laboratories, inc, USA) 24 h before LPC injection, then 0.5 μg DT every third day until 10 days after LPC injection. Control mice were treated with PBS injection. Flow cytometry in spleens and blood were used to confirm Tregs depletion. …

  • … To induce hair cell damage, DT (List Biological Laboratories, Inc., Campbell, CA) at 25 or 50 ng/g was injected intramuscularly at 7 weeks of age (1-week post-tamoxifen) once a day on two days spaced 44-48 hours apart. We selected a DT dose for each experiment based on periodic tests of DT potency, which varied with each lot and diminished over time during storage at -20°C …
