  • EAU model development and disease grading

A 1:1 mixture containing 2 mg/ml interphotoreceptor retinoid-binding protein 1–20 (IRBP1 − 20, GiL Biochem, Shanghai, China) and complete Freund’s adjuvant (CFA, BD Difco, San Jose, CA, USA) with 2.5 mg/ml of Mycobacterium tuberculosis (BD Difco, San Jose, CA, USA) was subcutaneously injected into mice (0.2 ml per mouse) on day 0. After immunization with IRBP1 − 20, mice were intraperitoneally injected with 0.2 µg pertussis toxin (PTX, List Biological Laboratories, Campbell, CA, USA) dissolved in phosphate-buffered saline (0.1 ml per mouse) immediately. On day 2, PTX was again intraperitoneally injected into mice at the same dose [6]. On day 14, we conducted funduscopic examinations on the mice. …

Product: Diphtheria Toxin, Unnicked, from Corynebacterium diphtheriae

  • Tamoxifen and diphtheria toxin treatment

Tamoxifen (MedChemExpress) was dissolved in corn oil (20 mg ml−1) and heated at 37°C for 3 hours. To activate Cx3cr1CreERT2-mediated recombinase, each mouse was injected with Tamoxifen (100 mg/kg body weight) by oral gavage every three days, starting from Day 12 post-intraportal injection, as indicated in Fig. 2e. For proliferation tracing experiment, DreERT2-mediated recombinase was introduced by a single dose of tamoxifen gavage (100 mg/kg body weight) on Day 12 post-intraportal injection, as indicated in Fig. 3f. For KC specific depletion, DreERT2-mediated recombinase was introduced by 8 doses of tamoxifen gavage (2 mg per dose), as indicated in Fig. 5d. A single dose of 200 ng DT (List Laboratories) was intraperitoneally introduced one week after the last Tamoxifen injection. The cancer cells were inoculated into liver through portal vein one day after DT injection.

Product: Diphtheria Toxin CRM197, Mutant

  • Diphtheria Toxin (149A) was purchased from List Laboratories (CA, U.S.). …

… Treg depleted Rag 1-/- mouse model: 5 × 105 CD4+CD45RBhi T cells purified from the spleen and MLN of B6.129(Cg)-Foxp3/J donor mice were intraperitoneally injected into Rag 1-/- mice to construct a colitis model. During the experiment, 10 µg/kg DT was intraperitoneally injected into Rag 1-/- mice every 7 d from week 1, onward. B6.129(Cg)-Foxp3/J mice, which are Foxp3DTR knock-in mice. These mice contained a human DTR and GFP inserted downstream of the internal stop codon of the X-linked Foxp3 gene. Foxp3+ Tregs in these mice highly express DTR-GFP. Administration of DT to these mice resulted in the depletion of Treg cells in lymphoid tissues within 2 d post-injection. …
