  • Mice and EAE induction

We purchased C57BL/6, (C57BL/6xSJL) F1, 2D2-TCR-Tg, and DO11.10-TCR-Tg mice from the Jackson Laboratory. Mice were maintained in a specific pathogen–free facility and used under protocols approved by the National Institute of Allergy and Infectious Diseases (NIAID) Animal Care and Use Committee (protocol LISB-7E). EAE was induced by immunization of female C57BL/6 mice (10 to 12 weeks old) with 200 μl of emulsion containing 100 μg of MOGp35–55 and 400 μg of Mycobacterium tuberculosis extract H37 Ra (Difco) in Freund’s complete adjuvant (FCA) as previously described (73). At 4 and 24 hours after immunization, mice received 200 ng of pertussis toxin (List Biological Laboratories) intraperitoneally. To induce RR-EAE in (C57BL/6xSJL) F1 mice, we immunized the female F1 mice with 200 μl of emulsion containing 200 μg of MOGp35–55 and 400 μg of M. tuberculosis extract H37 Ra (Difco) in FCA. At 4 and 24 hours after immunization, mice received 75 ng of pertussis toxin intraperitoneally. EAE was scored by the methods previously described (74, 75). …

2.5 EAE model establishment and treatment

A total of 50 mice were randomly divided into 5 groups (10 mice/group) as follows:
control group (CON), EAE model group (MOD), SJP low-dose group (SJP-L), SJP high-dose group (SJP-H), and positive control Prednisone acetate tablets group (PAT). The experimental autoimmune encephalomyelitis model was established as described previously(Shaw et al., 2011; Stromnes and Goverman, 2006). All mice were injected subcutaneously with an antigen-mixed emulsion (200 μg of MOG35-55 emulsified in a 1:1 ratio with CFA containing 4 mg/mL inactivated MTB) on day 0, followed by intraperitoneal injection of 500 ng PTX on days 0 and 2 to induce the EAE. …

  • 2.3 Induction of antibody-augmented MOG35-55 EAE

Active EAE was induced in 8-10 weeks old mice. MOG peptide 35-55 (MOG35-55, 100 µg; Institute of Medical Immunology, Charité Berlin, Germany) emulsified in complete Freund’s adjuvant (CFA) was injected subcutaneously (s.c.) under anesthesia (50 mg/kg Ketamine; Ketanest®, Zurich, Switzerland and 1 mg/kg medetomidine; Dormitor®, Provet, Lyssach, Switzerland). After 30 minutes, mice were awakened by a s.c. antagonist injection (Atipamezol 1.25 mg/kg; Antisedan®, Provet), hydrated with 300 µl sodium chloride 0.9% solution and observed until fully recovered. Zero and two days post immunization (dpi), 200 ng pertussis toxin (List Biological Laboratories, Campbell, CA, USA) was injected intraperitoneally (i.p.). …
