  • PT-specific lymphocytes and cytokine assessment

    At the respective timepoints, additional heparin tubes (12mL) were collected from children that were conveniently selected for assessment of their CMI responses against PT. CD3+ CD3+ CD4+ and CD3+CD8+ PT-specific lymphoblasts were measured using a specialized Flowcytometric Assay that detects the Specific CMI response in Activated whole blood(FASCIA). Similar to previous protocols [11, 16-18], whole blood was diluted 1/10 in complete RPMI medium (supplemented with 50 μg/mL gentamycine, 2 mM L-glutamine, MEM non84 essential AA, Na pyruvate, 2B-mercapthoethanol and Fetal Bovine Serum) and stimulated with 5 μg/mL PT antigen (heat-inactivated at 96°C for 20 minutes, Bordetella pertussis strain (native toxin), List Biological.
