  • Peanut sensitization, ButM treatment and challenge

SPF C3H/HeN mice were treated with 0.45 mg of vancomycin in 0.1 ml volume by intragastric gavage for 7 d pre-weaning and then with 200 mg l−1 vancomycin in the drinking water throughout the remainder of the sensitization protocol. Age- and sex-matched 3-week-old littermates were sensitized weekly by intragastric gavage with defatted, in-house-made peanut extract prepared from unsalted roasted peanuts (Hampton Farms, Severn, NC) and cholera toxin (CT) (List Biologicals) as previously described25,45. Sensitization began at weaning and continued for 4 weeks. Before each sensitization, the mice were fasted for 4–5 h and then given 200 ml 0.2 M sodium bicarbonate to neutralize stomach acids. After 30 min, the mice received 6 mg of peanut extract and 10 μg of cholera toxin (CT) in 150 ml PBS by intragastric gavage. …

  • 2.3. EAE induction and evaluation

EAE was induced using methods in a previous study [23]. Briefly, 200 μg of murine myelin oligodendrocyte glycoprotein (MOG) (GL Biochem, Shanghai, China) was emulsified in complete Freund’s adjuvant (CFA) [18], which consisted of 5 mg/mL Mycobacterium tuberculosis (BD, San Jose, CA, USA) and incomplete Freund’s adjuvant (Sigma-Aldrich, St. Louis, MO, USA). After which, the mice were injected with MOG/CFA subcutaneously on the left and right retroauricular spots and flank on day 0. Pertussis toxin (200 ng, List Biological Laboratories, CA, USA) was intraperitoneally injected into the ophthalmic vein on day 0 and day 2, respectively. The mice were monitored at appropriate times. …

    Cell death assay

Macrophages were plated onto 96-well plates and cultured for 12 h in RPMI medium containing 10% FBS. Macrophages were stimulated with the IAP antagonist, BV6 (1 μM), in the presence of the apoptosis inhibitor, zVAD (20 μM), the RIPK3 inhibitor, GSK’872 (5 μM), or both, as indicated, for the times indicated. To induce pyroptosis, macrophages were pretreated with LPS (10 ng/mL) for 4 h, then stimulated with nigericin (10 μM) for the indicated times. …
