  • Subjects and experimental outline:

In total, 40 mice (both genders) were used in this experiment with 20 each assigned to the control (Cre) and experimental (Cre+) groups. All subjects in both groups received one intraperitoneal injection (i.p.) of DT at a dose of 25 ng/g (List Biological Laboratories, Campbell, CA, USA) at postnatal day 21. …

  • Drug treatments:

…For fate-mapping studies in damaged utricles, tamoxifen (6 mg/40 g body weight, IP) was injected once on two consecutive days in 9-week-old Plp1-CreERT2:ROSA26tdTomato:Pou4f3DTR mice, followed one week later by two intramuscular (IM) injections of DT. Each injection was 25 ng/g body weight, and there was a 48 hr interval between injections. DT was purchased from…List Biological Laboratories, Inc. (Campbell, CA). The different tamoxifen doses produced similar labeling of utricular SCs. …

  • Diphtheria toxin treatment:

Diphtheria toxin (DT) was administered to DTR mice, genetically engineered to express the human DTR selectively in hair cells (Tong et al. 2015). A single 25 g/kg dose of DT (List Biological Laboratories, no. 150) was delivered via intramuscular injection to 4-wk-old DTR mice. Within 6 days after DT injection, DTR mice lose all of their hair cells and are completely deaf (Tong et al. 2015). After DT injection, DTR mice were allowed to survive for 2 wk before tissue collection.
