Immunization protocol and treatment of mice with antibiotics:
We anesthetized 6~8-week old age- and sex-matched mice with isoflurane and intranasally immunized the mice with 30 µl of PBS (Gibco) containing 30 µg of HSA and 100 ng of CT using a pipette and tip. For secondary immunization, mice were challenged with HSA (30 µg) and CT (100 ng) four weeks after the primary immunization.
For systemic immunization, mice were intraperitoneally injected with 100 µl of PBS containing 100 µg of HSA and various concentrations of CT.

Materials and Methods:
BoNT/A was radiolabelled by pretreatment with 2iminothiolane and incubation with 99mTc-gluconate. The labelled toxin 99mTc-BoNT/A was puried using size exclusion HPLC. Twenty-four female Wistar rats were evenly injected in the bladder wall with either 99mTc-/ (n = 12) or free 99mTc (n = 12). Four rats from each group were killed at 1, 3 and 6 h after injection, respectively. The bladder, L6-S1 spinal cord segment and L6-S1 dorsal root ganglia (DRG) were harvested and their radioactivity counted in a gamma scintillation detector. Results were calculated as % injected dose (I.D.) per gram of tissue. The paired t-test was used for comparison of means of 99mTc-/ radioactivity vs free 99mTc in the tissues of interest.

EAE induction and analysis:
EAE was induced in 8 to 12 week-old mice by immunization with 100 µg MOG p35-55 or rhMOG in complete Freund’s adjuvant containing 200 µg Mycobacterium tuberculosis H37Ra (Difco Laboratories, Detroit, MI) on day 0.
