Presentation matters: Impact of association of amphiphilic LPS with serum carrier proteins on innate immune signaling

Stromberg, LR;Mendez, HM;Kubicek-Sutherland, JZ;Graves, SW;Hengartner, NW;Mukundan, H;

Product: ULTRA PURE LPS from Escherichia coli O111:B4

  • Measuring the LPS induced response in IC-21 cells:

    A 12-plex mouse inflammatory cytokine multi-analyte ELISArray kit (Qiagen) paired with LPS O111:B4 (phenol extract, Sigma Aldrich) was used to determine how many hours of LPS exposure was required to produce a measurable cytokine response. Limit of detection for the ELISAarray 12-plex kit is stated by the manufacturer as 10 pg/mL for each given cytokine. For further in-depth examination we chose ULTRA PURE LPS O111:B4 (List Labs) to avoid contaminates often found in other LPS preparations. …

Real-Time Signaling Assays Demonstrate Somatostatin Agonist Bias for Ion Channel Regulation in Somatotroph Tumor Cells

Product: Pertussis Toxin from B. pertussis, Lyophilized in Buffer

Product: Unspecified List Labs LPS

  • Treatment of mice, and flow cytometry:

    Four-week-old male C57BL/6 mice were intraperitoneally (i.p.) injected with 25g of LPS (List Biological Laboratories, Inc., Campbell, CA) …

    Author did not specify which List Labs LPS product was utilized in their research.  List Labs provides the following LPS products:  https://listlabs.com/product-information/lipopolysaccharides/

Product: Toxin A from Clostridium difficile

  • Toxin Activity Inhibition Assay:

    … The cytotoxic titer was defined as the reciprocal of the highest dilution that produced rounding in 80% of Vero cells for each sample. Vero cells treated with purified C. difficile toxins (A and B) and antitoxin (List Biological Labs, 152C and 155C; TechLabs, T5000) were used as controls. A test cytotoxicity assay was run prior to assays to ensure that the 2-AI molecules did not affect the cytoskeleton of Vero cells at the tested concentrations.

Product: Pertussis Toxin from B. pertussis, Lyophilized in Buffer

  • Pertussis Toxin Preparation:

    Lyophilized pertussis toxin (List Biological) was reconstituted with distilled water. The reconstitute was then further diluted to a concentration of 2500 ng/ml then loaded into 1ml syringes for IP injection (Stromnes and Goverman, 2006).

    EAE Induction:

    Mice were shaved dorsally from the base of the tail to the middle of the back rostrally. Subcutaneous (SC) injections of MOG35-55/CFA emulsion were performed on day 0 and day 7. Each mouse received 4 subcutaneous (SC) injections of 0.05 ml at separate locations on the rump, yielding a total of 0.2 ml per mouse. Injections sites were arranged rostro-caudally, two on either side of the dorsal midline. A single IP injection 0.1ml of pertussis toxin was administered at a concentration of 2500 ng/ml on day 0 and day 2.

    Author did not specify which List Labs Pertussis Toxin was utilized. List Labs provides the following Pertussis Toxin products:
