Cholera toxin immunisation:
TCR/ mice were sublethally irradiated (500 rad) and reconstituted with B6 or Rorc/ bone morrow. Before immunization with cholera toxin mice were deprived of food for 2 h and then given 0.25 ml of a solution containing eight parts HBSS and two parts 7.5% sodium bicarbonate by oral gavage (o.g.) to neutralize stomach acidity. After 30 min, mice were o.g. immunized with 25 µg cholera toxin (List Biological Laboratories, USA). Concentrations of antibodies specific for cholera toxin were determined by ELISA with cholera toxin (List Biological Laboratories) as the capture agent.
