Product: Cholera Toxin (AZIDE-FREE) from Vibrio cholerae

  • Primary human cell cultures:

Human skin samples were obtained as anonymised surgical waste, typically from abdominoplasty or mammoplasty. Briefly, skin biopsies were minced and trypsinized (0.05% trypsin/0.01%EDTA) at 37C for 4 hr. Cells were collected every 30 min, plated (2.5  104/cm2) on lethally irradiated 3T3-J2 cells (2.4 X 104/cm2) and cultured in 5% CO2 and humidified atmosphere in keratinocyte growth medium (KGM): DMEM and and Hams F12 media (3:1 mixture) containing Fetal Bovine Serum (FBS) (10%), insulin (5 g/ml), adenine (0.18 mM), hydrocortisone (0.4 g/ml), cholera toxin (CT, 0.1 nM), …

Product: LPS from Escherichia coli O111:B4

  • Experimental Procedure:

On P10, cross-fostered ferret kits were randomized to receive an intraperitoneal 5 mg/kg dose of LPS (from Escherichia coli O111:B4, List Biological, CA, USA) or saline vehicle, before being returned to their jills for 4 h. LPS-injected animals were subsequently placed in a humidified chamber within a water bath, and underwent consecutive hypoxia, hyperoxia, and hypoxia (60 min at 9% O2, 120 min at 60% O2, and 30 min at 9% O2). …
