Western blot

To detect PT production by Western blot, to a 1.0 mL aliquot of culture supernatant, ¼ volume of 100% TCA was added, mixed, and the sample was held on ice for 1 h. Samples were centrifuged at 16,000 × g for 30 min at 4 °C and the supernatant was removed. The TCA-precipitated protein pellet was resuspended in 120 µL 1 × NuPAGE SDS Sample Buffer (Invitrogen) in the presence of 1 × NuPAGE Reducing Agent (Invitrogen) and denatured at 90 °C for 10 min. The assay was run using cryopreserved human PBMCs from a single healthy donor and were allowed to recover for 24 hours before use. On day 0, we stained human PBMCs with cell proliferation tracing dye (excitation: 488 nm and emission: 530/30 nm) and washed 3 times before plating into a 96-well plate at 2 million/mL (50 μL/well).
