Pasteurella Multocida Toxin

Pasteurella multocida toxin (PMT) is produced by native strains of Pasteurella multocida. Purified Pasteurella multocida toxin has a molecular weight of approximately 146 kDa, corresponding to 1,285 amino acids. This toxin can affect the signal transduction pathway of many cells by selective transient activation of a G-protein known as Gqα. Pasteurella multocida toxin has been reported to also activate Rho kinase through a Gqα independent pathway.

Pasteurella multocida toxin activates the Gqα subunit that stimulates phosphatidyl-inositol-specific phospholipase C-B1. Phospholipase C cleaves phosphatidyl-inositol producing secondary messenger compounds which affect several intracellular events such as the mobilization of calcium pools and protein phosphorylation. Pasteurella multocida toxin can also stimulate Rho kinase directly by a Gqα independent pathway. Some events that are reported to occur downstream in this signal transduction cascade are induction of stress fiber formation and stimulation of focal adhesion assembly through tyrosine phosphorylation of the focal adhesion kinase p125 FAK and paxillin.

Additional intracellular events that have been demonstrated to be caused by Pasteurella multocida toxin include increase in endothelial permeability by stimulation of Rho kinase and myosin light chain phosphatase. On a cellular scale, PMT stimulates Xenopus oocytes, inhibits some potassium currents in CHO cells, and is a potent mitogen of several types of cells including fibroblasts.
