  • Retrograde Labeling of Phrenic Motoneurons:

    Intrapleural injection of cholera toxin subunit B (CTB) was used to retrogradely label phrenic motoneurons in immunohistochemical and laser capture microdissection experiments. This technique was previously validated for this purpose and labels only phrenic motoneurons in the cervical spinal cord [28]. Briefly, 20 µl of a 0.2% solution of either Alexa Fluor 488-conjugated CTB (Molecular Probes; Life Technologies, Grand Island, NY) or unconjugated CTB (List Biological Laboratories, Campbell, CA) was injected using a Hamilton syringe into the pleural space of the right and left sides of the chest between the 7th and 8th ribs at 3 days before the terminal experiment.
