  • Retrograde Labelling and Perfusion:

    Bulbospinal neurons in the ventral medulla were retrogradely labelled with CTB (List Biological Laboratories; Campbell CA). Rats were anesthetized with isoflurane (3% for induction, 1.52% for maintenance). After removal of the spinous process of the T8 vertebra, a laminectomy was performed at T9. A small part of the rostral portion of the T10 vertebra was removed, as needed, to improve access to the spinal cord. The tip of a glass micropipette was targeted to the dorsal root entry zone (~0.51 mm lateral to midline), inserted to a depth of 1 mm and 3 60 nl of CTB was injected into each side of the cord (6 injections total). CTB injections were spaced between 0.2 and 1 mm apart in order to avoid damaging surface blood vessels. Three days after tracer injection, rats were anesthetized and perfused through the heart with DMEM/F12 tissue culture medium (D-8900; Sigma Chemical, St Louis, MO, USA) followed by 4% formaldehyde in 0.1M phosphate buffer, pH 7.4. Three untreated male Sprague-Dawley rats without CTB injections in T10 were also perfused. Brains and spinal cords in their vertebral columns were post-fixed in the same fixative for 3 days and washed in several changes of phosphate buffer. …
